Apple will spread the the details of iOS 5 at the WWDC 2011 in June. The release of iOS 5 is expected to happen in September along with the release of the iPhone 5. According to iPhone App developer FutureTap (via 9to5Mac), Apple is testing apps on iPhone 4 and first generation iPads running iOS 5.
